This article discusses the use of free DNA removal with MGCs Aspergillus Assays to ensure the assay is only amplifying DNA From live cells.
Yes you can, with a few important recommendations from MGC.
Using Grim Reefer when running the Aspergillus Assays with the SenSATIVAx Mag Bead based DNA Purification
Historically, MGC did not believe the GR option was necessary with microbial tests that are presence/absence and require an enrichment step since the purpose of enrichment is to increase the abundance of live cells.
However after listening to feedback from several of our customers that want the option to use GR with the Aspergillus assay, we created a supplemental guide for users to follow if running with Grim Reefer is desired. This process will require the samples to be enriched for a full 48 hours. The use of Grim Reefer with the Aspergillus Assays is not part of MGCs current AOAC PTM validation we will not be added. However we are including it in our AOAC validation for Aspergillus with the use of MaGiC Lysis - see below.
If your lab has any concerns with the use of Grim Reefer in conjunction with the Aspergillus assays, we recommend performing an internal validation. For help designing this validation please reach out to MGCs Support Team. Support@medicinalgenomics.com
Using Grim Reefer when running the Aspergillus Assays with the MaGiC Lysis DNA Isolation
The MaGiC lysis with both the 2-Color and 5-Color Aspergillus is available for purchase and is currently going through the process to gain AOAC approval. There will eventually be an option to use Grim Reefer Free DNA removal for this assay but will require a 48 hour enrichment.
