What do I do if my flower samples have no HEX? Can I spike in synthetic cannabis DNA during DNA purification for flower samples?

For cannabis flower samples which are not expected to contain adequate plant DNA (shake, keif, trim, and potentially remediated samples) synthetic cannabis DNA can be added during purification to help to ensure internal control recovery.

How to spike SCCG into purification process for cannabis flower samples:

1. Prepare a fresh SCCG positive control dilution of 1:50,000 and keep on ice until use.


2. Total Yeast and Mold Assay, Total Aerobic Count Assay, Total Coliform/Entero Assay, BTGN Assay:

After addition of lysis buffer, vortexing, and incubation, add 2.5 uL of the 1:50,000 SCCG dilution followed by another vortex.  Proceed with the next spin down and bead based DNA purification.

3. Presence/Absence assays:

After addition of lysis buffer, vortexing, and incubation, add 10 ul of the 1:50,000 SCCG dilution followed by another vortex.  Proceed with spinning the sample down and bead based purification.
