The new qPCR Master Kit v3 contains additional enzyme and is compatible with all pass/fail assays.
The Medicinal Genomics qPCR Master Kit v3 was developed and released in 2020 in conjunction with the PathoSEEK® 5-Color Aspergillus Multiplex assays. The qPCR Master Kit v3 can be used with any pass/fail assay without any change in protocol.
The only change with the qPCR Master Kit v3 is an increase in the amount of Taq DNA polymerase enzyme. The qPCR Master Kit v3 will eventually replace the qPCR Master Kit v2 across all enumeration assay protocols, but this transition will be gradual as we update SOPs, validation data, and standard curves for all assays.
Do any assays require the use of qPCR Master Kit v3?
At this time, only the 5-Color Aspergillus Multiplex assays for the Bio-Rad and Agilent Real-Time PCR platforms require the new qPCR Master Kit v3. Multiplexing five targets on five unique fluorophores requires more enzyme for the reaction.
No other assays require the use of the new qPCR Master Kit v3 at this time.

Can I use qPCR Master Kit v3 with any enumeration assays?
No. Using the new qPCR Master Kit v3 for any enumeration (total count) assays will require a new standard curve to be used. Since there is additional Taq enzyme in each reaction, the Cq/CFU conversion equation may shift for a given assay.
We are currently in the process of updating our validation and SOPs of our total count assays so that qPCR Master Kit v3 will be required. We plan to make these announcements and changes in Q2 of 2021.
