What's new with the Pseudomonas and Staphylococcus Version 2 Assay?

January, 2025. Medicinal Genomics released a new Staphylococcus and Pseudomonas multiplex assay that detects each organism in one reaction on separate optical channels. This includes a new User Guide and Validation document.

PathoSEEK® P. aeruginosa & S. aureus Multiplex Detection Assay v2 with MaGiC Lysis Kit (P/N 420526)

The PathoSEEK® P. aeruginosa & S. aureus Multiplex Detection Assay v2 with MaGiC Lysis Kit (PN 420526) will differentiate between Staphylococcus and Pseudomonas in one qPCR reaction.  Staphylococcus aureus is detected on the FAM channel and Pseudomonas aeruginosa is detected on the Cy5 channel.

MaGiC Lysis reagents and Amplification Mix will be included with the assay in the kit.

Version 1 Assays

In the previous version of the Staph/Pseudo Multiplex assay (PN 420138), both organisms were detected using the FAM channel. This limitation required the use of "singleplex" assays for additional DNA analysis to determine whether S. aureus or P. aeruginosa was present when a FAM signal was detected.

The Staph/Pseudo Multiplex assay (PN 420138), along with the individual Staph and Pseudo singleplex assays (P/Ns 420136 and 420137), will be discontinued later this year.

Validation

The new assay kit (PN 420526) has been validated for use with MGC's MaGiC Lysis, which offers a faster and more streamlined process compared to the previous SenSATIVAx method. MaGiC Lysis employs a straightforward boil preparation followed by qPCR, simplifying the workflow. For detailed instructions and validation information, please refer to the User Guide and Validation document linked here.

Compatibility

The new v2 assay and amplification mix are compatible with the SenSATIVAx DNA Purification kit. If the preferred DNA Isolation method is SenSATIVAx, the Staph and Pseudo Multiplex v1 Assay Kit (with Amplification Mix) can be ordered without MaGiC Lysis. SenSATIVAx will need to be ordered separately. Please contact support@medicinalgenomics.com if you have any questions.
