Why can't I use Grim Reefer with the Total Yeast and Mold Assay using TLP?

Grim Reefer and TLP are not compatible with one another.

In the Total Yeast and Mold assay the TLP is designed to lyse yeast cells.  If the Grim Reefer is added to the mixture after the TLP, the DNA released from the yeast cells will be digested by the Grim Reefer enzyme.  Grim Reefer is deactivated by the addition of the MGC lysis buffer which inhibits the function of TLP.  Therefore, if we inactivate the Grim Reefer with lysis buffer before addition of TLP, the TLP will not function as intended.
