What's new with FemINDICAtor® Version 3?
We have combined the new FemINDICAtor® Assay v3 with Amplification Mix in a "kit" format to simplify ordering along with other method updates.
We are pleased to announce the release of our new FemINDICAtor® Cannabis Sex Detection Assay Kit v3. This integrated "kit" combines the qPCR assay and Amplification Mix, supplying 200 reactions. This streamlined format simplifies ordering and reduces errors by ensuring all necessary components are used correctly and simultaneously.
Updates include the following enhancements:
- Kitted Format: The assay and amplification mix are now provided in a complete kit format.
- New Version 3 Assay: This updated assay features an optimized internal cannabis control.
- Validation: The assay has been formally validated with the specific Amplification Mix.
- Instrument Compatibility: Validation testing confirms compatibility with the Bio Molecular Systems Mic Thermal Cycler.
- Optimized Protocol: The Instructions For Use specify 20 µL reactions as the recommended volume to achieve optimal results across all sample quality types.
Improved Target Accuracy and Expanded Thermal Cycler Optimization
FemINDICAtor® v3 assay has been optimized for use on additional thermal cycler platforms, including the Biomolecular Systems Mic, by targeting a more ubiquitous cannabis gene for the internal control. This change reduces the need for reruns because of poor internal control amplification. When paired with the Mic, qPCR data review may be semi-automated with limited user intervention, improving workflow efficiency and consistency.
Protocol Update and Recommended Best Practices
Medicinal Genomics strongly encourages adoption of the FemINDICAtor® v3 Kit and adherence to the updated instructions for use. The v3 method was developed to deliver greater consistency and robustness across multiple qPCR platforms.
As part of these improvements, 20 µL reaction volumes are recommended as best practice. The increased reaction volume improves tolerance to PCR inhibitors and delivers more consistent performance across a broader range of sample qualities, extraction conditions, and qPCR instruments.
While 10 µL reaction volumes will continue to be supported, users should be aware that reduced volumes are more sensitive to PCR inhibitors. 10 uL reactions may not be advisable for samples with higher levels of background material or potential PCR inhibitors like foliar sprays.
Continued Support for Prior Versions
The previous FemINDICAtor® method is not obsolete and is still supported by Medicinal Genomics. Customers may continue to use the v2 assay (while supplies last) and associated protocols, including 10 µL reaction volumes, where applicable. However, Medicinal Genomics strongly encourages adoption of the FemINDICAtor® v3 assay and adherence to the updated instructions for use.
